Characterization of Microbial Community in Cold-Chain Hairtail Fish by High-Throughput Sequencing Technology

Abstract: High-throughput sequencing was used to analyze the microbial communities in the muscle samples of hairtail fish to study their diversity and dynamic changes during cold-chain circulation. The results showed that the richness and diversity of the microbial community in hairtail fish had a transient decline in 0–24 h and decreased after the first rise during 24–216 h. The diversity and richness of bacteria in cold-chain hairtail fish reached the maximum at 168 h. The Shannon and Simpson diversity indices of the … Show more

“…The hairtail ( Trichiurus lepturus ), a highly prized marine fish species, is widely distributed across the eastern Pacific Ocean and along the coastlines of various regions in China [ 1 ]. Among China’s four major commercial fish species, the hairtail is notably rich in vitamins, minerals, polyunsaturated fatty acids, proteins, and essential amino acids, such as eicosatetraenoic acid and docosahexaenoic acid [ 2 , 3 ]. Consequently, several hairtail products, including air-dried hairtail [ 4 ], vinasse hairtail fillets [ 5 ], and deep-fried hairtail fillets [ 6 ], have been developed.…”

“…In order to improve the above problem, many related scholars are committed to studying the quality change in hairtail during logistics and transportation and its maintenance methods. Scholars analyzed microbial communities in hairtail muscle samples using the Illumina MiSeq high-throughput DNA sequencing platform [15]. This method can be used to study the diversity and dynamics of microbial communities in hairtail during the cold chain.…”

“…It is therefore important to characterize the microbiota of this fish in different stages of post-harvest to identify the sources of contamination. Owing to the limitations of plate-based morphological and biochemical characterization, culture-independent high-throughput sequencing (HTS) has been used to trace down the potential sources of microbial contamination in post-harvest fishes and food products (Bledsoe et al 2016;Antunes-Rohling et al 2019;Xing et al 2021). In this study, we aimed to identify and compare the skin and gut microbiota of live, frozen and marketed rohu fish samples, with the aid of culture-independent quantitative real-time polymerase chain reaction (RT-PCR) and 16S rRNA gene sequencing technology.…”
