Characterization of Organic Phosphorus in Lake Sediments by Sequential Fractionation and Enzymatic Hydrolysis

Abstract: The role of sediment-bound organic phosphorus (Po) on lake eutrophication was studied using sequential extraction and enzymatic hydrolysis by collecting sediments from Dianchi Lake, China. Bioavailable Po species including labile monoester P, diester P, and phytate-like P were identified in the sequential extractions by H2O, NaHCO3, and NaOH. For the H2O-Po, 36.7% (average) was labile monoester P, 14.8% was diester P, and 69.9% was phytate-like P. In NaHCO3-Po, 19.9% was labile monoester P, 17.5% was diester P… Show more

“…Also concentrations of NaOH and EDTA in these mixtures are varied slightly among studies depending upon their objectives. Recoveries ranged from 57 to 90 % with 0.25 MNaOH-50 mM EDTA extraction of soil (Makarov et al 2002) and sediment (Zhu et al 2013a;Zhang et al 2014). In this study, the optimal extraction of P o from aquatic macrophytes and algae was achieved by use of 0.5 M NaOH-25 mM EDTA ( Table 2).…”

“…However, P o and condensed P from aquatic macrophytes were that orthophosphate monoesters (90.3 %, 819 mgkg −1 ) > orthophosphate diester (5.5 %, 50 mg kg − 1 ) > pyrophosphate (4.2 %, 38 mg kg − 1 ) (Zhu et al 2013a), the average of 18 sediments in lake Dianchi d C/N, C/P, N/P refers to the mass ratio (Table 4). The composition of P o and condensed P in the debris from aquatic macrophytes and algae was different, though orthophosphate monoesters were the major component of P o from both.…”

“…Although organic P can constitute 30-80 % of the P in lake sediments (Williams and Mayer 1972;Boström et al 1982), it has been relatively under-studied compared to inorganic phosphate (Mitchell and Baldwin 2005). In particular, organic P occurs in a variety of forms that behave differently in the aquatic environment (McKelvie 2005), so information on the relative abundance of P compounds in sediments is of importance for understanding P dynamics in lakes (Reitzel et al 2007;Turner and Weckström 2009;Zhu et al 2013). However, organic P is conventionally determined by sequential fractionation and detection by molybdate colorimetry Condron and Newman 2011).…”

“…Recent studies have determined the chemical nature of sediment organic P by alkaline extraction and solution 31 P nuclear magnetic resonance (NMR) spectroscopy, which classifies organic P into a series of compounds and functional groups, including phosphomonoesters, phosphodiesters (e.g., DNA, lipids), and phosphonates (e.g., Carman et al 2002;Ahlgren et al 2005;Reitzel et al 2007;Zhu et al 2013). The technique also identifies inorganic phosphates, including orthophosphate, pyrophosphate, and polyphosphate (e.g., Hupfer et al 2004).…”
