“…Genetic transformation in European plum (Prunus domestica L.) has been established through regeneration of the cotyledon and hypocotyl sections of mature embryos (Mante et al, 1989;Scorza et al, 1994), immature embryos and embryonic axes (Tian et al, 2007a;Srinivasan et al, 2012;Petri et al, 2012: Wang et al, 2013. Medium supplemented with thidiazuron (TDZ) 7.5 μM in combination with indole-3-butyric acid (IBA) 0.25 μM has been used for shoot regeneration, with a transformation effi ciency of 1.2% (Scorza et al, 1994;González-Padilla et al, 2003).…”

“…Shoots were induced from hypocotyl segments of mature seeds on medium containing TDZ, 6-benzylaminopurine (BA) and kinetin. TDZ medium was the most eff ective for shoot regeneration, with an effi ciency of 30% of explants forming shoots; the shoot induction response was genotype-dependent (Tian et al, 2007a). Urtubia et al (2008), using a modified protocol for regeneration of European plum (Scorza et al, 1994), obtained 11% and 19% shoot regeneration from hypocotyl slices in the Japanese plum cultivars "Angeleno" and "Larry Anne", respectively.…”

“…Particularly, in the common plum (Prunus domestica L.), a commercially important stone fruit tree, the rooting efficiency is low, despite the attempts directed towards higher effectiveness of the process (choice of the auxin, root induction procedure, one-or multi-step culturing, treatment duration etc.) (Gonzales Padilla et al 2003;Tian et al 2007). Despite numerous studies on regeneration potential of the common plum, the process of adventitious rhizogenesis itself has not been investigated extensively in this important fruit tree.…”

“…communication). Our previous studies indicate that these plums have low response to in vitro regeneration (Tian et al 2006) and the genetic transformation efficiency is also low (Tian et al 2009). Plum fruits, two weeks prior to maturation, were collected from plum trees in Vineland, Ontario, Canada.…”

“…Indeed, European plum genetic transformation has only been successful in a few laboratories. Tian et al (2006) evaluated in vitro regeneration of European plum germplasms and varieties adapted to high latitude and developed genetic transformation technology for these types of plum plants via hopcotyl regeneration (Tian et al 2009). Nevertheless, at this time, wide and practical use of plum genetic transformation technology in many other laboratories and studies is still not feasible and is difficult.…”
