Characterization of Riboflavin-modified Dentin Collagen Matrix

Abstract: Crosslinking is considered a possible approach to increasing the mechanical and structural stability and biodegradation resistance of the dentin collagen matrix. The aim of this study was to investigate the mechanical and chemical variations and collagen degradation resistance associated with crosslinking of the dentin collagen matrix with UVA-activated riboflavin. Dentin collagen matrix specimens were treated with 0.1 and 1% riboflavin for 2 min and photo-activated with 7 mW/cm(2) UVA (368 nm) for 2 min. The … Show more

“…In contrast to the current results, some previous studies stated that dentin surface preparation with RF/UVA increased the µTBS values immediately and after water storage (9, 30). They attributed this increase in bond strength to the stiffness of dentin collagen matrix (9,30,32), upstanding of collagen fibrils and enhanced resin infiltration following cross-linking (30), decreased activity of MMPs due to increased cross linking of dentin collagen (9,29) and direct cross linking of MMPs (9). The difference between the current results and those of previous studies may be explained as follows:…”

“…Due to the biocompatibility and cross linking of dentin collagen matrix, this technique may be used as a method of dentin surface preparation or as an optimizer of adhesive systems (29). This study aimed to assess the efficacy of riboflavin activated with light (by a light curing unit) for collagen cross linking and optimizing the bond strength of an etch and rinse adhesive system to dentin.…”

“…The most likely reason for significant improvement in the bulk mechanical properties of non-demineralized dentin is the formation of covalent crosslinks in the dentin collagen matrix, which was detected by Raman analysis. Recently, UVA-RF was found to crosslink the collagen matrix, and to increase the mechanical properties of demineralized dentin (7). In this study, the dentin specimens were etched with 37% phosphoric acid to dissolve the mineral components and expose the collagen matrix to UVA-RF directly.…”

“…UVA-activated riboflavin (UVA-RF) has been considered to be a successful treatment strategy for progressive keratoconus because of its consistent stabilizing effect in corneal collagen crosslinking (4). Recently, UVA-RF has been shown to enhance the mechanical properties and mechanical stability of dentin collagen matrix (6,7). However, these studies focused on completely demineralized dentin collagen.…”
