Characterization of the inhibitory roles of RFRP3, the mammalian ortholog of GnIH, in the control of gonadotropin secretion in the rat: in vivo and in vitro studies

Abstract: van Noort PI, Tena-Sempere M. Characterization of the inhibitory roles of RFRP3, the mammalian ortholog of GnIH, in the control of gonadotropin secretion in the rat: in vivo and in vitro studies. Am J Physiol Endocrinol Metab 299: E39 -E46, 2010. First published April 27, 2010; doi:10.1152/ajpendo.00108.2010.-RF-amide related peptides (RFRP), as putative mammalian orthologs of the avian gonadotropin-inhibitory hormone (GnIH), have been proposed as key regulators of gonadotropin secretion in higher vertebrates.… Show more

“…As in the ovine, in vitro and in vivo data of a study of the bovine also strongly suggest a pituitary site of action of GnIH [31]. In spite of the lack of GnIH fibres in the external zone of the median eminence in the rat, compelling data for a pituitary site of action has been provided for this species [32]. …”

“…ICV administration of GnIH reduced phosphorylated P450arom in the POA in the short term compared with control birds . The action of GnIH on neuroestrogen synthesis in the POA was abolished by concomitant administration of RF9 (Pineda et al, 2010;Simonin et al, 2006), a potent antagonist of GnIH receptor, or fadrozole (Steele et al, 1987;Wade et al, 1994), an inhibitor of P450arom . Accordingly, it is considered that GnIH stimulates neuroestrogen synthesis in the POA by activating P450arom through dephosphorylation after binding to GnIH receptor in P450arom cells.…”

“…In addition to its hypothalamic expression, GnIHR expression has been reported in the pituitary (Ubuka et al 2009b) and the gonads (McGuire & Bentley 2010). The inhibitory effect of GnIH on reproductive hormones secretion can be blocked by the GnIHR antagonist RF9 (Pineda et al 2010a,b, Rizwan et al 2012.…”
