Characterization of the Mineral Fraction Associated to Extracellular Polymeric Substances (EPS) in Anaerobic Granular Sludges

Abstract: The extracellular polymeric substances (EPS) extracted from four anaerobic granular sludges contain an important mineral fraction (20-77% of the EPS dry weight). The composition of the mineral fraction of EPS depends strongly of the extraction method applied and to a lesser extend of the origin of the anaerobic sludge. Centrifugation, sonication, and heating extraction procedures yield a similar mineral composition. However, extraction using a cationic exchange resin (CER) leads to an increase of the Na(+) con… Show more

“…Fig. 1 illustrates the detailed procedures of each extraction process, which were carried out according to methods developed by previous studies (Liu and Fang, 2002;d'Abzac et al, 2010;Adav and Lee, 2008). To investigate the changes of sludge characteristics resulting from EPS extraction, the sludge samples (10 mL) were obtained and analyzed before centrifugation at 20,000 × g during the extraction process.…”

“…Extracellular polymeric substances (EPS) are high molecular weight compounds secreted by microorganisms into their environment (Sheng et al, 2010) and include proteins, polysaccharides, humic substances, deoxyribonucleic acids (DNA), lipids, and uronic acid Liu and Fang, 2002;Sheng et al, 2010;d'Abzac et al, 2010). EPS, an important composition of activated sludge Nielsen et al, 1996), plays an important role in biological wastewater treatment (Wilen et al, 2003;Neyens et al, 2004;Wang et al, 2005;Long et al, 2009).…”

“…Chemical extraction protocols have also shown to contaminate the EPS extracts by impurities or carrying over of the extractant itself 26 . According to some studies, the molecular movement is enhanced, which accelerates the EPS dissolution 29 , so heating is the most effective method for EPS extraction due to its minimal cell lysis and more EPS extracted 30,31 .…”
