Characterization of the pJB12 Plasmid from Pseudomonas aeruginosa Reveals Tn 6352 , a Novel Putative Transposon Associated with Mobilization of the bla VIM-2 -Harboring In58 Integron

Abstract: The bla VIM-2 -carrying In58 integron has been linked to a chromosomal location in different bacterial species, including Pseudomonas aeruginosa. This work reports the first fully sequenced In58-harboring plasmid, which is significantly different from the two previously identified bla VIM-2 -carrying plasmids in P. aeruginosa. bla VIM-2 might have been acquired by transposition of Tn6352, a novel transposon composed of the In58 and ISPa17 elements. The recognition of similar inverted repeat (IR) sites by ISPa1… Show more

“…One C. freundii (Cfr-12, ST129) carried the bla VIM-2 gene in a 30 kb plasmid. It had almost 100% identity with pJB12 (KX889311), detected in four P. aeruginosa from inpatients of two different hospitals in Portugal 20 , 21 . pJB12 has the bla VIM-2 -carrying In58 integron immersed in Tn 6352 , a transposon composed of the In58 and IS Pa17 elements.…”

“…A large plasmid carrying the rare bla SIM-2 gene cassette in a class 1 In/Tn (320) and a smaller plasmid carrying bla KPC-2 , different from the P-6/U plasmids with bla KPC mentioned above (321), were both sequenced recently. A group of related plasmids that carry a cassette-borne bla VIM-1 or bla VIM-2 carbapenemase gene in different class 1 In/Tn structures were sequenced (318,(322)(323)(324). They are also related to TNCP23, a region carrying a class 1 integron bounded by two copies of IS6100 (IS6 family) that seems to correspond to a plasmid inserted into the larger plasmid pKLC102, which is itself known to integrate as a genomic island (see below) (325).…”

“…Plasmids have played an important role in acquisition of resistance genes in P. aeruginosa, including carbapenemases (93)(94)(95)(96)(97), highlighting the importance of understanding constraints to plasmid transfer in this pathogen. Systematic surveys of the abundance of plasmids in P. aeruginosa are lacking, in part due to the challenges of identifying plasmids from short read assemblies, but it is clear that plasmids are much less abundant than ICE, as P. aeruginosa genomes typically contain multiple ICE (Fig.…”
