Characterization of the presumptive sarcoma cells in primary MSV tumors

Abstract: Presumptive sarcoma cells have been isolated from primary MSV tumors induced in normal, immunosuppressed (ALG) and athymic nude mice. These cells were atypical in appearance, induced tumors in secondary hosts and expressed the viral antigens gp70, p30 and p12. In vitro growth characteristics of the MSV cells were tested in a variety of assays defining cell transformation. None of these indicated that the virus-positive cells were transformed. Sex chromosome marker studies were carried out to determine if secon… Show more

“…These results strongly suggest that reinfection of the second host is a very important mechanism in the successful "transplantation" of the lesion, and thus supports the notion of Simons (1970) (Becker & Haskill, 1980b) (Zinkernagel, 1979). Also, we wanted to re-examine our previous conclusion (Becker & Klein, 1980) that cytotoxic T cells are not generated in response to MSV in the A/Sn strain.…”

“…Although these macrophage cell fractions were inactive in the 16h 51Cr-release or microcytotoxicity experiments (Fig. 2) we have been able to show that these cells exert both cytostatic and cytolytic effects against the autologous "sarcoma" cells in 48h assays (Becker & Haskill, 1980b).…”

“…The macrophages could not be purified from the "sarcoma" cells by adherence, since some of the "sarcoma" cells were highly adherent (Becker and Haskill, 1980a).…”

“…Mesenchymal cells, which are subsequently seen to proliferate in the local lesion and give rise to fibroblasts, are not typical of this process and may be characteristic of sarcomas. Yet the great majority of these tumors regress, and unlike other tumors they do not appear to develop by clonal proliferation, but may spread by reinfection of adjacent normal cells by virus, present at the local site in high concentration (Becker and Haskill, 1980). Accordingly regression could be thought to be due to elimination of the virus and cessation of virus production.…”
