Characterization of the Subunit Structure of the Ribonucleic Acid Genome of Influenza Virus

Abstract: Ribonucleic acid extracted from influenza virus was labeled at the 3' termini with 3H and analyzed by polyacrylamide gel electrophoresis. Influenza virus was found to contain a minimum of seven and possibly as many as 10 polynucleotide chains, most of which appear to terminate at the 3' end in uridine.

“…2. "NTM" (nonterminal materials) denotes the position where proteins, carbohydrates, and mononucleotides elute (4,8). Label appearing there indicates either that the sample contained traces of proteins or carbohydrates, or that a low level of labeling of internal nucleotides occurred.…”

“…The specificity of the labeling and analytical procedures was monitored with R17 phage RNA (bottom panel, Fig. 2), which is known to terminate at its 3' end in AOH (3,8 for 40 hr at 10 ma/tube in 4% polyacrylamide-bis acrylamide gels (14 by 0.7 cm in diameter) by using a running buffer composed of0.04 M tris(hydroxymethyl)aminomethane, pH 7.4, 0.001 M ethylenethiaminetetraacetic acid, and 0.005 M sodium acetate. Direction of migration is from left to right.…”

“…The single-stranded RNA genome (vRNA) of influenza virus is segmented (4,18,28). The ts mutants of influenza WSN virus grown in MDBK cells fall into at least seven complementation-recombination groups (30,31), and the working hypothesis is that there is a one-to-one correspondence between a complementationrecombination group and a vRNA segment.…”

“…Moreover, from the analysis of recombinants between two strains of influenza A viruses, the protein corresponding to each of the eight genes has been identified (24,26,31,32). Structural studies indicate that each RNA segment possesses a unique nucleotide sequence (6, 14, 18) but that all or most of the segments contain pppA at the 5' end (39) and uridine at the 3' end (17).…”
