Characterization of verruciform xanthoma by in situ hybridization and immunohistochemistry

Abstract: Verruciform xanthoma (VX) is a rare, benign lesion, mainly found in the oral mucosa. Histologically and ultrastructurally, the lesion is characteristic and well defined. However, the etiology of the lesion remains unclear. The purpose of the present study was to elaborate upon the pathogenesis of VX by evaluation of an additional series of oral examples for human papillomaviruses (HPV), using immunohistochemistry and in situ hybridization, and to further characterize the cellular components of VX immunohistoch… Show more

“…The etiology is unknown, and most cases are not related to human papilloma virus (HPV) infection. The presence of HPV in the epithelial cells has been demonstrated in only two reported lesions in the oral mucosa [15] and scrotum [16].…”

“…The size of oral VX lesions normally ranges between 0.2 to 2.0 cm 3,4,[16][17] with exception of a large lesion measuring 4.0 x 1.5 cm reported by Graff et al 19 . With regard to appearance, Neville et al, described that this may vary from "white and verruciform" to "red and ulcerated".…”

“…The treatment of choice was to perform excisional biopsy of the two lesions, with excellent prognosis up to the present date. Treatment was performed by surgical excision, in which recurrence is extremely rare [16][17] . An interesting finding in the present case was the fact that the patient presented two oral lesions.…”

“…1,10,11,17,18 Evaluation for HPV in VX has been studied and has included types 6, 7, 11, 12, 16 to 19, 31, 33, 35, 37, 44 to 47, 53, 54, and 58. 6,13,[21][22][23] While most studies failed to show evidence of HPV, 1 study 13 demonstrated by immunohistochemical analysis a positive reaction in the upper epidermal keratinocytes, which was confirmed by electron microscopy and by PCR, which also detected HPV-DNA 6a. Another investigation 22 demonstrated by immunohistochemical analysis a positive reaction with polyclonal antisera against papilloma virus.…”
