Chemical Approach for the Study of the ‘Kissing Complex’ of Moloney murine leukaemia Virus

Abstract: The replication of Moloney murine leukaemia virus relies on the formation of a stable homodimeric kissing complex of a GACG tetraloop interacting through only two C · G base pairs flanked of 5'adjacent unpaired adenosines A9. Previous NMR investigations of a model stem loop 1 has not permitted to reveal the origin of this interaction. Therefore, with the aim of deeper comprehension of the phenomena, the model sequence 10 was prepared where position 9 has been substituted for a nucleoside offering a wider p-sta… Show more

“…Due to availability of labeled acetone, we decided to introduce the third ring with [D 5 ]bromoacetone. [17] We explain the unexpected product mixture with a ketoenol tautomerization of the bromoacetone under the reaction conditions. In this way, the deuterium atoms at the methylene position of bromoacetone are exchanged with hydrogen atoms, which are likely donated by the guanosine compound 5.…”

“…We considered the “third ring” to be a suitable and economic site to introduce deuterium labels. Due to availability of labeled acetone, we decided to introduce the third ring with [D 5 ]bromoacetone 17. Consequently, bromine was treated with [D 6 ]acetone in the presence of [D 1 ]acetic acid to yield, after careful distillation, pure [D 5 ]bromoacetone.…”
