“…Hemoglobin from bovine blood (Sigma H2500), comprising mostly metHb, was used as received. Na2S • 9H2O (Aldrich, 431648) was used as H2S donor, which dissociates in solution and reacts with H + to yield HS − , H2S and a trace of S 2− [26]. We use the term 'sulfide' to encompass the total mixture of H2S, HS − and S 2− .…”

“…Na2S • 9H2O was washed in distilled H2O and then dissolved in H2O to give a stock solution of~100 mM, which was determined by the absorbance (ABS) at λmax 232 nm, ε = 7700. The concentration of polysulfides in this stock was <10 μM as judged by UV-VIS spectrophotometry [26]. This stock solution was always prepared fresh right before the experiment, kept in sealed vials with minimal headspace and used within 4 hours.…”

“…[16][17][18] The endogenous H 2 S can generate H 2 S n by reacting with ROS. [8][9][10][19][20][21] However, H 2 S n derivatives are proposed to be direct antioxidants against ROS in cells. 22,23 There is no doubt that H 2 S n possess their own bio-functions, but the evidence is limited owing to the lack of accurate and sensitive detection methods.…”

“…A more sensitive method is based on monobromobimane in which two monobromobimane molecules form the stable sulfide dibimane in the presence of H 2 S. Sulfide dibimane can be separated by reverse-phase chromatography and detected by a fluorescence detector. Now, fluorescent probes and sulfide selective electrodes are extensively used however, with different sensitivity and reliability (148). As yet, H 2 S measurements are difficult, with variable reliability, thereby complicating studies on the role of H 2 S and its use as therapeutic target in various diseases, including diabetes-associated vascular disease.…”
