Cacao (Theobroma cacao L.) is a crop of globally importance on whose production around 20 million people depend directly. The crop is threatened due to the incidence of diseases caused by phytopathogenic fungi such as Moniliophthora roreri, causing losses of more than 80 % of the annual production. For this reason, in this study, a natural product such as the essential oil (EO) obtained from Siparuna guianensis was evaluated as a control alternative. The chemical composition of the EO, as well as the antifungal effect in vitro and in vivo on M. roreri, the causal agent of moniliasis frosty pod rot, were determined in the cacao pods located in a commercial plantation in southern Huila, Colombia. 28 compounds were detected, predominating: D-Germacrene (26.5 %), (E)-nerolidol (21.5 %), β-caryophyllene (9.3 %), elemol (8.0 %), bicyclogermacrene (7.5 %), δ-elemene (3.5 %), β-elemene (3.0 %), and α-pineno (2.4 %). At a concentration of 1000 μg ml-1 of EO in vitro, an inhibitory effect of 98 % on the mycelial growth of M. roreri was obtained. In the in vivo test at concentrations of 1000 μg ml-1 and 750 μg ml-1, a direct relationship in the decrease of the incidence, external gravity, and internal gravity of M. roreri in cocoa pods was observed, using EO as fungicide. It was concluded that S. guianensis EO represents a great potential for the control of frosty pod rot.

BACKGROUNDThe shortcomings of synthetic pesticides are becoming increasingly evident. In the pursuit of looking for alternative, environmentally friendly pesticides, the potential use of two naturally occurring sesquiterpenes, α‐bisabolol and bisabolol oxide A, as bactericides or acaricides, was investigated against three gram‐positive and three gram‐negative bacteria, and the eggs and larvae of the cattle tick Hyalomma scupense, respectively. The primary targeted site of synthetic insecticides is acetylcholinesterase (AChE), so the inhibitory activity of both sesquiterpenes was evaluated. Then, molecular docking and molecular mechanics generalized Born surface area (MM‐GBSA) studies were performed to gain insight into their binding mode in the AChE active site.RESULTSBisabolol oxide A showed good activity against Staphylococcus aureus, Escherichia coli, and Salmonella enteritidis with inhibition zones ranging from 16 to 34 mm and minimal inhibitory concentration (CIM) of 3.125 mg mL−1. Ovicidal and larvicidal assays revealed promising activity of α‐bisabolol and bisabolol oxide A against H. scupense, with bisabolol oxide A being more effective against larval ticks with lethal concentration (LC50) value of 0.78%. Both compounds also displayed potent anti‐AChE activity with inhibition concentration (IC50) values of 37.09 and 28.14 μg mL−1, respectively. Furthermore, α‐bisabolol and bisabolol oxide A exhibited good and comparable docking scores (−7.289 and −7.139 Kcal mol−1, respectively) and were found to accommodate in the active‐site gorge of AChE via hydrogen bonding and hydrophobic interactions.CONCLUSIONBisabolol oxide A and, to a lesser degree, α‐bisabolol are active against bacteria and ectoparasites and may represent an economical and sustainable alternative to toxic synthetic pesticides to control pathogens. © 2024 Society of Chemical Industry.

This study aimed to determine the repellent and insecticidal activity of four essential oils (EOs) from plants collected in the Chocó rain forest, Colombia, against T. castaneum. Conventional hydrodistillation was used to obtain the EOs. The repellent and insecticidal activities were evaluated by the preference area and gas dispersion methods, respectively. Statistical differences (p<0.05) were determined by applying a student’s t-test. EOs of Siparuna guianensis, S. conica, Piper marginatum, and Nectandra acutifoliashowed excellent repellent properties as the main findings, highlighting S. conicaEO with 84% repellency (1μL/cm2), while P. marginatumshowed to be bioactive to the dose of 500 μL/mL (72 h), inducing mortality of 100% of the exposed population. In conclusion, the results evidenced the repellent properties of the EOs evaluated against T. castaneum, which allows us to conclude that these plant species are potential natural sources producing bio-repellents that contribute to the integrated control of T. castaneum.
