Chemical synthesis and biological activity of rat INSL3

Abstract: The recently identified protein, insulin 3 (INSL3), has structural features that make it a bona fide member of the insulin superfamily. Its predicted amino acid sequence contains the classic two-peptide chain (A- and B-) structure with conserved cysteine residues that results in a disulphide bond disposition identical to that of insulin. Recently, the generation of insl3 knockout mice has demonstrated that testicular descent is blocked due to the failure of a specific ligament, the gubernaculum, to develop. Th… Show more

“…Ovine and rat INSL3 were chemically synthesized and characterized as described (7,9). Human INSL3 and biotinylated ovine INSL3 were prepared similarly with the ovine INSL3 containing a single biotin molecule on the N terminus of the A chain.…”

“…Male mice mutant for INSL3 exhibit bilateral abdominal cryptorchidism (2,3) whereas female mice overexpressing INSL3 showed ovary descent and displayed bilateral inguinal hernia (4). Although INSL3 binds to gubernacular homogenates (5,6) and induces growth of rat gubernaculum in whole organ cultures (7), the exact nature of the INSL3 receptor is unknown.…”

“…The native peptides of INSL4 through 6 have not been isolated. There is currently no other structural information on any other relaxin peptide, although circular dichroism analysis of INSL3 generally indicates that it has a tertiary structure similar to that of relaxin (Bü llesbach and Schwabe, 1995;Dawson et al, 1999;Smith et al, 2001). …”

“…Overexpression of the INSL3 protein in female mice causes the ovaries to descend into the inguinal region due to an overdeveloped gubernaculum (9). Synthetic INSL3 will induce the growth of the gubernaculum in whole organ cultures (10). Hence the peptide is essential for the transabdominal phase of testicular descent where the gubernaculum proliferates and results in the migration of the testis to the inguinal region.…”
