Chemiluminescence analysis of the activity of Pecten maximua hemocytes stimulated with zymosan and host-specific Rickettsiales-like organisms

Abstract: ABSTRACT. During a study of Rickettsiales-like organisms (RLO) in Pecten maximus gill, histological observation of free RLO in hemolymph led us to consider interactions between the pathogen and the hemocytes. Firstly, the production of oxygen radicals by hemocytes was established using a luminolenhanced chemiluminescence method with zymosan as stimulant. The effects of several inh~bitors of oxidative metabolism were investigated. The internalization of RLO into hemocytes was demonstrated by electron microscopy… Show more

“…Hemocytes from many bivalve species are known to be able to produce ROS. Oxidative mechanisms were reported in the oysters C. virginica, C. gigas, C. ariakensis and O. edulis (Larson et al, 1989;Bachère et al, 1991;Hégaret et al, 2003b;Lambert et al, 2003;Goedken and DeGuise, 2004;Labreuche et al, 2006;Donaghy et al, 2009), the mussels M. edulis and M. galloprovincialis (Pipe, 1992;Carballal et al, 1997b;Ordás et al, 2000b) and the scallops P. maximus and C. farreri (Le Gall et al, 1991;Chen et al, 2007a,b). In clams, few data are currently available and contradictory results between species were reported.…”

“…In vertebrates, phagocytosis of foreign material or chemical stimulation was shown to induce an increase in ROS production (Bedard and Krause, 2007;Nauseef, 2008;Segal, 2008). In bivalve species, phagocytosis of zymosan particles increased oxidative mechanisms in the oysters C. gigas , C. virginica (Hégaret et al, 2003b), O. edulis , the clam species S. patula (Greger et al, 1995), M. mercenaria (Buggé et al, 2007), as well as in the pectinid P. maximus (Le Gall et al, 1991). Activation with chemicals such as phorbol myristate acetate (PMA) also increased ROS production in C. virginica (Goedken and DeGuise, 2004) whereas, PMA exposure resulted in a decrease in ROS in C. gigas hemocytes .…”

“…Adaptations for avoiding or bypassing the haemocytic oxidative microbicidal system have been demonstrated in several pathogens, using in vitro host-pathogen models with isolated Ž . specific pathogens Hervio et al, 1989;Le Gall et al, 1991;Volety and Chu, 1995 . In crustaceans, the demonstration of respiratory burst is quite recent. Using the Ž .…”

“…The oxidative activity in molluscan hemocytes has been shown to be valuable in defining host-defen sive events (Adema et al, 1991;Anderson, 1994;Greger et al, 1995) and hemocytepathogen interactive modulation (Le Gall et al, 1991;Anderson et al, 1992b;Anderson, 1997, 1998).…”
