Chemiluminescence of Acanthamoeba castellanii

Abstract: 1. Chemiluminescence of Acanthomoeba castellanii in the presence of O2 was of similar intensity in organisms harvested early or late during exponential growth [when cyanide (1 mM) stimulates or inhibits respiration respectively]. 2. Cyanide (up to 1.5 mM) stimulated photoemission in both types of organism by 250--300 photons/s per 10(7) cells above the value observed under aerobic conditions. 3. 'Dibromothymoquinone' (2,5-dibromo-6-isopropyl-3-methyl-p-benzoquinone) (up to 80 microM) further increased chemilum… Show more

“…The O 2 -dependence of H 2 O 2 generation was also demonstrated in this system. In a similar experiment, we monitored low-level chemiluminescence (Lloyd et al, 1979), characteristic of singlet O 2 (Fig. 3c).…”

“…Absorbance and light scattering measurements (Park et al, 1997) were done in a Beckmann DV 7500 UV/ visible spectrophotometer. Oxygen consumption at controlled low O 2 concentrations was in a stirred reaction vessel fitted with a Radiometer (Copenhagen) O 2 electrode in a Hitachi 557 doublebeam spectrophotometer (Degn et al, 1980) or used in an EMI single photon counting luminometer fitted with a cooled (220 uC) low-background photomultiplier tube (Lloyd et al, 1979;Biagini et al, 2001). Gas mixtures were made using pre-purified N 2 ; O 2 and solubility tables were used to calculate dissolved O 2 (Wilhelm et al, 1977).…”

“…Hydroperoxides were used as 0. [12][13][14][15][16][17] Amol/min caused only a moderate increase in chemiluminescence, but higher rates, ,mol/min, produced a marked augmentation of light emission (Fig. 2).…”

“…6n5i10' cells ml − ") were exposed to low levels of dissolved O # in a continuously stirred reactor (200 r.p.m.) fitted with an observation window for photon counting (Lloyd et al, 1979(Lloyd et al, , 1985. The reactor was open for gas exchange from a mobile gas phase, the composition of which was controlled using a gas mixer ; dissolved O # was monitored using a Radiometer electrode.…”
