Chemoenzymatic Synthesis of Campylobacter jejuni Lipo-oligosaccharide Core Domains to Examine Guillain–Barré Syndrome Serum Antibody Specificities

Abstract: Guillain−Barrésyndrome is often caused by Campylobacter jejuni infection that has induced antibodies to the lipo-oligosaccharide (LOS) that cross-react with gangliosides at peripheral nerves causing polyneuropathy. To examine fine specificities of anti-ganglioside antibodies and develop a more robust platform for diagnosis and disease monitoring, we developed a chemoenzymatic approach that provided an unprecedented panel of oligosaccharides composed of the innercore of the LOS of C. jejuni extended by various… Show more

“…Glycan array binding analysis of the rHAs was carried out as described here. 50,51 Briefly, recombinant hexahistidine-tagged HA was precomplexed with a mouse anti-his Alexa 647 antibody (Abcam) and goat - anti-mouse Alexa 647 antibodies. This was done in 50 μL PBS-T (phosphate-buffered saline with 0.1% Tween-20) in a 4:2:1 molar ratio, incubated for 15 minutes on ice, and the applied on the array for 90 minutes in a humidified chamber.…”

“…Some examples that are of special interest for glycan mimicry are gangliosides and poly-LacNAc repeats that were synthesized through automated synthesis in combination with enzymes ( Li et al. 2019 ) or the LOS of C. jejuni that was synthesized through chemoenzymatic synthesis ( Li, Wolfert, et al. 2020 ).…”

“…Campylobacter jejuni is a gram-negative organism that is a common cause of bacterial diarrhea in developed and developing countries and is known as an infectious agent before the onset of neurological symptoms of Guillain-Barre syndrome. It is thought that due to the molecular similarity of the membrane antigens of this bacterium with peripheral nerve gangliosides, antibodies made against the bacterial membrane antigen cause damage to the peripheral nerves [12,13]. In addition to causing clinical symptoms, Guillain-Barre syndrome may be mistaken with poliomyelitis and since polio is close to eradication, all cases of acute flaccid paralysis, including Guillain-Barre syndrome, are being carefully examined [14].…”
