Chemoprevention of dietary digitoflavone on colitis-associated colon tumorigenesis through inducing Nrf2 signaling pathway and inhibition of inflammation

Abstract: BackgroundNuclear factor-erythroid 2-related factor 2 (Nrf2) has emerged as a novel target for the prevention of colorectal cancer (CRC). Many chemopreventive compounds associated with Nrf2 activation are effective in preclinical systems and many on-going clinical trials are showing promising findings. In present study we evaluated the cytoprotective effect and chemopreventive properties of dietary digitoflavone.MethodA cell based Antioxidant Response Element (ARE)-driven luciferase reporter system was applied… Show more

“…Nrf2 plays a critical role in protecting against inflammation-associated CRC, supporting and extending a published report showing that Nrf2 knockout increases the susceptibility to inflammation-associated aberrant crypt foci [75]. Activation of Nrf2 signaling resulted in suppression of colitisassociated CRC [76][77][78][79]. We showed that 10% cocoa activates Nrf2 (Figs.…”

“…In order to confirm that the mechanism of enzyme-II induction is via ARE activation [38], we evaluated the Nrf2 nuclear translocation ability, using the 1,2,5-oxadiazole (3) as the model compound. Therefore, to further investigate effects of (3) on the Nrf2/ARE activation, we examined the subcellular location of Nrf2 in Hepa-1c1c7 cells after 1,2,5-oxadiazole treatment.…”

“…The Nrf2 signaling pathway is also activated during inflammation and negatively regulates inflammatory responses. Inflammation can induce the expression of Nrf2 downstream genes (9), and the Nrf2 pathway cross talks with NF-B and other inflammatory signaling pathways to repress the inflammatory response (10).…”
