Chemoprevention of polyp recurrence with curcumin followed by silibinin in a case of multiple colorectal adenomas

Abstract: Chemoprevention is a practical approach to reduce the risk of various cancers including colorectal cancer (CRC). The goal is to reduce the incidence of pre-neoplastic adenomatous polyps and prevent its progression to CRC. Curcumin and silibinin prevent intestinal polyp formation in mice. Curcumin sensitizes silymarin to exert synergistic anticancer activity in colon cancer cells. Patients presenting with multiple colorectal adenomatous polyps (MCRA) have a high lifetime risk for CRC. We present a 57-year-old m… Show more

“…The role of CUR was investigated in the prevention of cancer and treatment of pre-invasive tumors, reporting some preliminary results [55,56,57]. Oral and topic formulations were proposed, tailoring high-risk patients with CUR-based pharmacological interventions and tracking the reduction of pre-invasive lesions when exposed to CUR and related compounds.…”

“…Surprisingly, a difference in polyp size and number between placebo and curcumin was not found. Furthermore, a case report [66] of a patient with 54 polyps at index colonoscopy, with all genetic tests negative for genetic polyposis demonstrated an interesting result. Indeed, after the removal of about 40 polyps, the patient was given curcumin 400 mg/day for three months.…”

“…Astonishingly, no variation in polyp numbers along with sizes amongst placebo as well as curcumin was observed. Moreover, a case report [38], in a patient with 54 polyps at index colonoscopy with all the genetic investigation negative regarding genetic polyposis illustrated intriguingly that subsequent to approximately 40 polyps surgical elimination, administration of 400mg curcuminfor3mths followed by silibinin. Following 2yrs just3 polyps were picked up at the follow up colonoscopy.…”
