Chemopreventive Strategies for Inflammation-Related Carcinogenesis: Current Status and Future Direction

Abstract: A sustained and chronically-inflamed environment is characterized by the presence of heterogeneous inflammatory cellular components, including neutrophils, macrophages, lymphocytes and fibroblasts. These infiltrated cells produce growth stimulating mediators (inflammatory cytokines and growth factors), chemotactic factors (chemokines) and genotoxic substances (reactive oxygen species and nitrogen oxide) and induce DNA damage and methylation. Therefore, chronic inflammation serves as an intrinsic niche for carc… Show more

“…In addition to the balance of Th1/Th2 ratio, AOM/DSS treatment causes colon tissue damage and it induces cytokine productions by neutrophil, monocyte, and macrophage. 42 After colon tissue damage, like T and B cells, these cells produce cytokines including TNF-α, IL-1β, and IL-6, causing inflammatory cell infiltration. Therefore, total cytokines are produced from T cell, B cell, and these three other cells.…”

“…ROS participate in neutrophils’ cytotoxic activity against cancer cells [ 35 , 74 ]. Nevertheless, enhanced ROS production by neutrophils limits the NK cell–mediated antibody-dependent cytotoxicity against leukemic cells during anti-CD20 treatment, induces DNA mutations and genotoxicity, and favors drug resistance and systemic T-cell and NK-cell dysfunction [ 77 , 78 , 79 ]. Collectively, these findings suggest that neutrophil-derived ROS can exert a protumorigenic action.…”

“…Studies to date have focused on the roles of DNA mutations as drivers of carcinogenesis. Nevertheless, many studies have established that DNA methylation changes are associated with chronic inflammation and cancer progression (Kanda et al, 2017;Zaware and Zhou, 2017). Although the presence of a small number of highfrequency CNVs in the field effect complicates the interpretation, our data suggest that early epigenetic changes involving DNA methylation could in fact cause the subsequent genomic alterations.…”
