Chemotherapy of Arthritis Induced in Rats by Mycobacterial Adjuvant

Abstract: Arthritis induced in rats by mycobacterial adjuvant has been used for the study of compounds of known value in the treatment of rheumatoid arthritis in man. The development of the arthritic syndrome in treated and control rats was followed by measuring the changes in foot thickness of both hind-feet with a micrometer. This method allowed the effect of anti-inflammatory compounds to be expressed quantitatively. Anti-inflammatory activity was readily observed in certain steroids, pyrazolidines, salicylates and s… Show more

“…In the rat, degranulation of mast cells entails the release of histamine and 5-hydroxytryptamine and, in this species, the peptide is, therefore, strongly inflammatory. We found, however, that in both the rat-paw oedema test 1191 and in adjuvant arthritis in rats [20] the peptide also shows powerful anti-inflammatory activity [18]. Whether these two (apparently opposite) biological activities are related is, at the moment by no means clear [18,21].…”

“…EEAB was found to significantly decrease the humoral immune response plausibly by inhibiting the acute inflammatory reaction by reducing vascular permeability and /or inhibiting other mediators. The secondary lesions are reported to presume due to a delayed hypersensitivity reaction [10] and EEAB exerted a marked effect on this. The aforementioned observations were also supported by the X ray pictures showing significant reduction in the inflammation, by EEAB, comparable to aspirin.…”

“…The effect of EEAB at 20 mg/kg was found to be slightly more than that of aspirin after 6 h of treatment (Table 1). Table 1 also summarized the effect of EEAB (5,10,20 mg/ kg) and aspirin on formaldehyde induced arthritis. EEAB significantly suppressed the joint oedema when compared with control between day 2 and day 10 post formaldehyde Table 2 showed the time course of oedema due to CFA and inhibition rate after the administration of EEAB.…”
