Chitosan-Alginate Blended Nanoparticles as Carriers for the Transmucosal Delivery of Macromolecules

Abstract: Nanoparticles intended for use in the transmucosal delivery of macromolecules were prepared by the ionic gelation of chitosan (CS) hydrochloride with pentasodium tripolyphosphate (TPP) and concomitant complexation with sodium alginate (ALG). The incorporation of a small proportion of ALG of increasing molecular weight (M(w); from 4 to 74 kDa) into the nanoparticles led to a monotonic increase in colloidal size from ∼260 to ∼525 nm. This increase in size was regarded as a consequence of the formation of gradual… Show more

“…As can be observed in Figure 2, pullulan-based nanoparticles evidence similar structure and morphology, corresponding to spherical and compact nanoparticles. This morphology corresponds to that widely reported for many formulations of polysaccharide-based nanoparticles (Calvo et al, 1997b;Goycoolea et al, 2009;Oyarzun-Ampuero et al, 2009;Sarmento et al, 2006;TeijeiroOsorio et al, 2009;Zorzi et al, 2011).…”

“…Faktor lain yang lebih menguatkan dinding mikrokapsul adalah ditambahkannya STPP yang mendorong terjadinya reaksi crosslink antara kitosan dari NH 3 + dengan gugus fungsi P 3 O 5 -10 dari STPP. Penambahan alginat diharapkan terjadinya kompleksasi polielektronik dengan gugus fungsi -COO -dari alginat (Goycoolea et al, 2009;Schatz et al, 2004;Jayanudin et al, 2015).…”

“…Enabling the controlled release of the active ingredient from the formulation is the outstanding property of chitosan that makes it desirable to be used in many novel drug delivery systems. [28][29][30][31][32] Depending on the molecular weight of chitosan, the released amount of the active ingredient may easily be controlled. Our results are in good correlation with the literature findings about the effect of molecular weight on drug release from the pharmaceutical formulations.…”
