Chitosan-Coated-PLGA Nanoparticles Enhance the Antitumor and Antimigration Activity of Stattic – A STAT3 Dimerization Blocker

Abstract: The use of nanocarriers to improve the delivery and efficacy of antimetastatic agents is less explored when compared to cytotoxic agents. This study reports the entrapment of an antimetastatic Signal Transducer and Activator of Transcription 3 (STAT3) dimerization blocker, Stattic (S) into a chitosan-coated-poly(lactic-co-glycolic acid) (C-PLGA) nanocarrier and the improvement on the drug's physicochemical, in vitro and in vivo antimetastatic properties post entrapment. Methods: In vitro, physicochemical prope… Show more

“…CS as a coating material led to an increase in encapsulation efficiency of the NPs. In the literature, there are several studies claiming that CS enhances the entrapment efficiency of NP systems [ 46 48 ]. While CS coats the surface of the NP, it also allows for the adsorption of the drug molecules on the NP surface [ 49 ].…”

“…The enhancement of DOX+VIN/Nio/CS cytotoxicity is due to endocytosis, which allows more niosomes to be taken up into the cells. 29,30 Also, in Figure S1C,D, the toxicities of Nio and Nio/CS formulation were determined toward the SKBR3 cell line in 48 and 72 h, respectively. There is a significant decrease in cell viability of Nio/CS compared to the Nio at all concentrations in 48 and 72 h except 0.25 μg/mL in 48 h (p < 0.05).…”

“…In addition, chitosan nanoparticles have already been shown to be excellent inhibitors of bacteria, both Escherichia coli and Staphylococcus aureus [ 172 ]. Recent findings include the use of chitosan in the preparation of nanocarriers to enhance the antimetastatic properties of the entrapped drug in vitro and in vivo [ 173 ].…”
