Chloroquine Resistant Plasmodium falciparum in Nigeria: Relationship between pfcrt and pfmdr1 Polymorphisms, In-Vitro Resistance and Treatment Outcome

Abstract: This study was designed to evaluate the association between polymorphisms in pfcrt and pfmdr1 genes and in-vitro chloroquine (CQ) sensitivity in fresh isolates of P. falciparum and patients' treatment outcome. The modified schizont inhibition assay was used to determine in-vitro sensitivity of P. falciparum. Polymorphisms in pfcrt and pfmdr1 genes were detected using nested PCR and RFLP techniques in 84 P. falciparum isolates obtained from patients with acute uncomplicated malaria.Eighty five percent (71/84) a… Show more

“…These results are in accordance with previous studies in Kenya (17) and Nigeria (57). However, the 86Y mutation has been associated with a decrease in CQ susceptibility in some works (5).…”

“…The N86-K76 and 86Y-K76 haplotypes showed a lower CQ IC 50 , in concordance with previous work in Kenya (17,61). The additional Pfmdr1 86Y mutation significantly increased the in vitro susceptibility to CQ (P ϭ 0.0179) in Pfcrt 76T CQ-resistant parasites from Senegal, while the 86Y-76T haplotype was shown to be associated with in vitro CQ resistance in isolates from Nigeria (57).…”

“…Similar K76T pfcrt genotypes were observed in the parasite genomic DNA extracted from filter-paper blood samples and saliva samples. The high prevalence of the mutant pfcrt 76T allele in both saliva (57%) and blood (60%) specimens is consistent with previous reports [20][21][22] from genomic DNA extracted from blood sample.…”

“…Nested PCR with RFLP techniques are frequently used to detect polymorphisms in parasite genes that serve as molecular markers of antimalarial drug resistance. [20][21][22] This approach was used in saliva samples to detect K76T polymorphism on the pfcrt gene that is associated with chloroquine resistance. Similar K76T pfcrt genotypes were observed in the parasite genomic DNA extracted from filter-paper blood samples and saliva samples.…”
