“…We have previously demonstrated that the TPK1 promoter, but not the other PKA subunit promoters, is activated upon heat shock and osmostress (Pautasso and Rossi, 2014;Reca et al, 2020). We also demonstrate that, during exponential growth, the BCY1 promoter has the lowest activity of the four PKA subunit promoters and does not respond to thermal stress (Pautasso and Rossi, 2014).…”

“…Several levels of control acting simultaneously and in coordination allows the specificity of the cAMP-PKA signal transduction. Our previous results have demonstrated that the mechanisms involved in TPKs and BCY1 expression in response to stress or growth conditions are subunitspecific (Galello et al, 2017;Pautasso and Rossi, 2014;Reca et al, 2020).…”

“…As high PKA activity is linked to lower stress tolerance, it seems plausible that regulation of PKA subunits at the transcriptional level might represent a mechanism to fine-tune PKA activity in response to stress. Some work has been carried out to study the regulation of TPK1-3 and BCY1 during heat shock and has identified that the TPK1 isoform is specifically upregulated, while TPK2 and TPK3 show no such increase in expression [ 101 , 102 , 103 ]. Curiously, a recent study of Tpk1 regulation during recurrent heat shock has shown that although TPK1 mRNA becomes more abundant, it is translationally inhibited and stored in mRNA granules until the stress is removed ( Figure 4 ) [ 103 ].…”

“…During stress conditions, heat shock and osmostress, the TPK1 promoter is the only one of the PKA subunit promoters that is upregulated. This promoter presents three Author's personal copy positioned nucleosomes that are evicted upon heat stress or osmostress, in correlation with promoter activation and upregulation of TPK1 mRNA levels (Pautasso and Rossi 2014;Reca et al 2020). It was demonstrated that the kinase Rim15 and the transcription factors Msn2/4, Gis1, and Sok2 are involved in TPK1 upregulation during heat shock (Pautasso and Rossi 2014).…”
