Chromosome 21-derived hsa-miR-155-5p regulates mitochondrial biogenesis by targeting Mitochondrial Transcription Factor A (TFAM)

Abstract: The regulation of mitochondrial biogenesis is under the control of nuclear genes including the master mitochondrial transcription factor A (TFAM). Recent evidence suggests that the expression of TFAM is regulated by microRNAs (miRNAs) in various cellular contexts. Here, we show that hsa-miR-155-5p, a prominent miRNA encoded in chromosome 21, controls the expression of TFAM at the post-transcriptional level. In human fibroblasts derived from a diploid donor, downregulation of TFAM by hsa-miR-155-5p decreased mi… Show more

“…Mitochondrial TFA binds mitochondrial DNA and regulates mitochondrial transcription initiation, mtDNA copy number, packaging of mitochondrial DNA, and mitochondrial biogenesis. 41 , 42 , 58 61 Thus, HN enhances RPE protection from oxidative stress via increased mitochondrial biogenesis and bioenergetics and by inhibiting the translocation of Bax from cytosol to mitochondria and preventing cytochrome c release. 14 Effects of HN and its analogues on mitochondrial function have been described for human aortic endothelial cells, cardiac myoblasts, human lymphocytes, muscular TE671 cells, and neural SKN-MC cells.…”

“…TFAM can be regulated by microRNAs and observations of post-transcriptional modifications of TFAM expression are performed by miRNA 155-5p. A significant negative correlation between miR-155-5p and TFAM expression was found in patient cardiac samples [43]. Yamamoto et al had an interesting finding regarding skeletal muscle and mitochondrial biogenesis.…”

“…The MIR155 (originally, BIC) gene has been studied for roles in lymphoma (reviewed in Lawrie 2013; Faraoni et al 2009), in mitochondrial function, in AD and inflammation and in regulation of a number of neurologically relevant target genes (Quiñones-Lombraña and Blanco 2015; Song and Lee 2015; Bofill-De Ros et al 2015). The MIR99AHG, or MicroRNA 99A host gene, contains three microRNA genes within its introns: mir99a, let7c and 125b2.…”
