Chromosome 3 Status in Uveal Melanoma: A Comparison of Fluorescence In Situ Hybridization and Single-Nucleotide Polymorphism Array

Abstract: ,7-9 PURPOSE. To compare fluorescence in situ hybridization (FISH) using a centromeric probe for chromosome 3 (CEP3) and 3p26 locus-specific probe with single-nucleotide polymorphism array (SNP-A) analysis in the detection of high-risk uveal melanoma.METHODS. Fifty cases of uveal melanoma (28 males, 22 females) treated by enucleation between 2004 and 2010 were analyzed. Fresh tissue was used for FISH and SNP-A analysis. FISH was performed using a CEP3 and a 3p26 locus-specific probe. Tumor size, location, a… Show more

“…Moreover, the location (ciliary vs. choroidal) or initial height of the tumor also had no statistical correlation with the regression rates following radiation therapy. Our results are in agreement with some of the previously reported studies [19,21,23]. …”

“…The specimen was immediately taken to the genetics laboratory in special transport media. Evaluation of the specimen was performed by an accredited cytopathologist using the fluorescence in situ hybridization (FISH) technique [15,19]. The minimum number of cells required for cytogenetic analysis was 200 cells; a smaller number of cells was considered to be an inadequate sample.…”

“…94 79,[125][126][127] and singlenucleotide polymorphisms (SNPs). 109,[128][129][130][131][132] The above-mentioned chromosomal alterations in primary UM are clinically relevant because of their correlation with the risk of metastatic death. Chromosome 3 loss is associated with a reduction of the 5-year survival probability from approximately 100% to o50%.…”

“…This is confirmed by studies examining UM metastases using FISH, demonstrating that the disseminated tumour cells are characterised by chromosome 3 loss and 8q gain. 109,113 We are currently examining paired primary and metastatic UM using aSNP and functional assays with the aim of identifying signalling pathways that are aberrant within the metastases and, consequently, potential 'druggable targets'.…”
