Chronic injection of pansomatostatin agonist ODT8-SST differentially modulates food intake and decreases body weight gain in lean and diet-induced obese rats

Abstract: The aim of this study was to investigate the central actions of the stable pansomatostatin peptide agonist, ODT8-SST on body weight. ODT8-SST or vehicle was acutely (1 μg/rat) injected or chronically infused (5 μg/rat/d, 14d) intracerebroventricularly and daily food intake, body weight and composition were monitored. In lean rats, neither acute nor chronic ODT8-SST influenced daily food intake while body weight was reduced by 2.2% after acute injection and there was a 14g reduction of body weight gain after 14… Show more

“…Conscious rats were placed lightly restrained (Ͻ1 min) in the holder tube of the quantitative nuclear magnetic resonance analysis apparatus (ECHO MRI, 3-1 Composition Analyzer; 150 Echo Medical Systems, Houston, TX). The body composition (fat mass, lean mass, and total water) was measured as described before (53).…”

“…Body composition was assessed in conscious lightly restrained (ϳ1 min) rats using a quantitative nuclear magnetic resonance analysis apparatus (EchoMRI-700 Composition Analyzer; Echo Medical Systems, Houston, TX), as described before (49). Changes in fat mass, lean mass, and total water were determined by assessing body composition before and 2 wk after switching to standard or high-protein diets.…”

“…In all, this study adds to the growing literature interrogating the contributions of TN SST neurons to feeding behavior. Central SST (originally named growth hormone inhibiting hormone, GHIH, in the central nervous system, (Painson and Tannenbaum, 1991) had long been known to affect food intake through somatostatin receptor 2 (SSTR2; (Beranek et al, 1999; Campbell et al, 2017; Danguir, 1988; Lin et al, 1989; Andreas Stengel et al, 2010b, 2010a; A Stengel et al, 2010; Stengel et al, 2015, 2013, 2011; Tachibana et al, 2009). This effect was seemingly localized to the tuberal nucleus, when TN SST neurons were found to integrate into the melanocortin feeding system, though the effect of these neurons on feeding was attributed to γ-aminobutyric acid (GABA) release as opposed to direct SST effects (Luo et al, 2018).…”
