Chronodisruption: effects on reproduction, transgenerational health of offspring and epigenome

Abstract: The circadian system regulates the daily temporal organization in behavior and physiology, including neuroendocrine rhythms and reproduction. Modern life, however, increasingly impacts this complex biological system. Due to limitations of working with human subjects exposed to shiftwork schedules, most chronoregulation research has used rodent models. Recent publications in these model systems have emphasized the negative effects of circadian rhythm disruption on both female and male reproductive systems and f… Show more

“…The placenta exposes maternal melatonin to the fetus at daily rhythmic intervals, with low concentrations during the day and high concentrations at night ( 8 ). Disrupting this rhythm during pregnancy may lead to detrimental outcomes for the mother and the growing fetus in utero and in adult life ( 8 , 56 , 61 , 73 ). The development of the embryo, uterine implantation, placentation, and delivery may be regulated by the clock molecular machinery in the circadian rhythm ( 73 ).…”

“…Diurnal rhythms of sperm count, sperm motility, and sperm chromatin integrity have been found in rodents. There are also several studies which have investigated the diurnal variation of semen parameters in humans ( Ni et al, 2019 ; Sati, 2020 ). Variations in semen parameters were found across different time points in most of the studies, although a high-resolution sampling study is required to confidently profile the circadian pattern.…”

“…Exclusion criteria included requirements for publication in English and operational definitions of shiftwork and/or nightshift, with a focus on primary research papers over meta-analyses. Additionally, the scope of this review includes reproductive and fertility measures up to birth, but does not focus on the long-term influence of shiftwork on offspring, although it should be noted that there is evidence that shiftwork can exert generational effects [81]. Within this review, shiftwork was defined as exposure to any combination of evening shifts, night shifts, and/or rotating shifts.…”
