Chryseobacterium profundimaris sp. nov., a new member of the family Flavobacteriaceae isolated from deep-sea sediment

Abstract: A Gram-stain negative, strictly aerobic, rod-shaped, non-motile bacterium, designated strain DY46(T), was isolated from Atlantic Ocean sediment. The isolate was found to grow in medium containing 0-3.0 % (w/v) NaCl (optimally at 0-1.0 %), at 4-37 °C and pH 5.0-8.0. Chemotaxonomic analysis detected MK-6 as the sole isoprenoid quinone. The major fatty acids were identified iso-C15:0, iso-C17:0 3-OH, iso-C17:1 ω9c and summed feature 3 (comprising iso-C15:0 2-OH and/or C16:1 ω7c). The DNA G + C content was determi… Show more

“…The Biolog Gen III Microplate system was used to determine substrate utilization according to the manufacturer's manual. Hydrolysis of starch (1.0 %, w/v), gelatin (1.0 %, w/v), l-tyrosine (0.5 %, w/v), carboxymethyl-cellulose (CMC, 1.0 %, w/v) and Tween 20, 40, 60 and 80 (1.0 %, w/v) were examined according to Xu et al [42]. Nitrate and nitrite reduction was determined by Griess A solution (0.5 g p-aminobenzenesulfonic acid with 150 ml (10 %, v/v) acetic acid), Griess B solution (0.1 g α-naphthylamine with 150 ml (10 %, v/v) acetic acid and 20 ml distilled water) and diphenylamine solution (0.5 g diphenylamine with 100 ml concentrated sulfuric acid and 20 ml distilled water) following cultivation in liquid medium added with 0.5 % (w/v) nitrate or 0.01 % (w/v) nitrite for 5 days.…”

“…PHB granules was detected by fluorescence microscopy (DMi8; Leica) after staining with Nile blue [35]. Oxidase activity was tested by oxidation of p -aminodimethylaniline oxalate solution (1 %, w/v) [36], and catalase activity was identified from the formation of bubbles in the hydrogen peroxide solution (3 %, v/v) [37]. Hydrolysis of gelatin (1 %, w/v), starch (0.2 %, w/v), casein (100 g l −1 skimmed milk; BD), Tween 20, Tween 40 and Tween 80 (1 %, w/v) was examined according to Xu et al [37].…”

“…Growth at different pH (5.0-10.5 with 0.5 pH unit intervals) was determined in marine broth 2216 by adding appropriate biological buffers: MES (pH 5.0-6.0), PIPES (pH 6.5-7.5), Tricine (pH 8.0-8.5) and CAPSO (pH 9.0-10.5). The temperature range (4,10,15,20,25,30,35,37,40,45 and 50 °C) for growth was investigated in MB. Cell motility was tested using a semisolid agar medium (per litre: 5 g agar, 37.4 g marine broth 2216) [34].…”
