The effect of PRL on the diurnal changes in peripheral lymphocyte and granulocyte number, antisheep red blood cells, and natural anti-rabbit red blood cells serum agglutinins titre as well as plasma corticosterone concentration was examined in White Leghorn cockerels, immunized twice with sheep red blood cells. PRL was administered for six consecutive days at 4 or 8 h after light onset. Control birds were treated at the same times with hormone solvent alone. Immunized non-treated birds served as an additional control group. PRL injections influenced markedly the diurnal changes in all parameters examined. The effect of PRL administration on the diurnal changes in lymphocyte and granulocyte number and natural anti-rabbit red blood cells serum agglutinins consisted in elimination of the influence of solvent injections. In those cases where the solvent injections did not alter the pattern of the diurnal changes, i.e. in anti-sheep red blood cells serum agglutinins and in plasma corticosterone concentration, the pattern was modified by PRL injections. PRL administration affected also the correlations between the diurnal changes in plasma corticosterone concentration and those in lymphocyte number and anti-sheep red blood cells agglutinin titre. This suggests that the role of PRL in the regulation of the diurnal variations of immunity in chickens may be realized either directly, via its receptors in immune system or by its influence on plasma corticosterone concentration.

A seasonal variation in the proliferative response to mitogens and in the proportion of splenic lymphocyte subpopulations was found in mice housed in a constant environment. The lymphoproliferative responses to T-cell and B-cell mitogens reached maximum values in autumn and summer. Identification of lymphocyte subpopulations by flow cytometry demonstrated that the proportion of T cytotoxic-suppressor (Tcs) lymphocytes was significantly higher in autumn and summer than in spring and winter. However, the proportion of B lymphocytes was significantly lower in spring than in the other three seasons, whereas the proportions of T and T helper (Th) cells did not show any seasonal variation. On the other hand, we observed a significant correlation between the level of mitogenic responsiveness and the proportion of Tcs cells, but not between the former and the proportions of B, T or Th cells. These data suggest that the seasonal variation in murine lymphoproliferative responses may depend on the cyclic changes in the proportion of Tcs lymphocytes; these changes, in turn, may be predetermined by the inherent internal biological rhythms of the animal.

Evidence for a Direct Action of Melatonin on the Immune System

Pineal melatonin modulates the mammalian immune system. In vivo studies showed that melatonin enhanced the natural and acquired immunity while in vitro studies demonstrated its inhibitory influence. The mechanism of melatonin action on the immune system remains unknown. Actions through lymphokines or opioid release or via other endocrine changes have been proposed. In this paper, a direct action of melatonin on the lymphoid tissue is hypothesized. 2-[¹²⁵I]Iodomelatonin binding sites have been identified in the membrane homogenates of thymus, bursa of Fabricius and spleens of a number of birds and mammals. The bindings were stable, saturable, reversible, specific and of high affinity. The Bmax ranged from 0.6 to 3.9 fmol/mg protein. The Kd was in the physiological range of circulating melatonin levels, about 30–70 pmol/l. The binding sites in the primary lymphoid organs demonstrated diurnal variation in density, with higher levels found at the middle of the light period. However, those in the spleen did not vary with the time of the day. An age-dependent decrease in the density was also found in the chicken bursa of Fabricius. In addition, when the noctural melatonin secretion was suppressed by constant light exposure, the density of the binding sites increased in the guinea pig spleen. Immunosuppression with Cortisol injection in young ducks decreased the density of the melatonin binding sites in the thymus. The regulation of the binding characteristics by physiological variation in melatonin levels and/or immunological status of the animals provide evidence that these 2-[¹²⁵I]iodomelatonin binding sites in the lymphoid tissues may be physiologically significant and represent true melatonin receptors. The melatonin receptors in the lymphoid organs may be coupled to a G protein as Guanosine 5''-O-(3-thiotri-phosphate inhibited 2-[¹²⁵I]iodomelatonin binding in the spleen by increasing the Kd and decreasing the Bmax.
