Circular RNA Expression and Regulation Profiling in Testicular Tissues of Immature and Mature Wandong Cattle (Bos taurus)

Abstract: Wandong cattle are an autochthonous Chinese breed used extensively for beef production. The breed tolerates extreme weather conditions and raw feed and is resistant to tick-borne diseases. However, the genetic basis of testis development and sperm production as well as breeding management is not well established in local cattle. Therefore, improving the reproductive efficiency of bulls via genetic selection is crucial as a single bull can breed thousands of cows through artificial insemination (AI). Testis dev… Show more

“…In the analysis of circRNA expression characteristics of testicular samples from 6-month-old bulls, a total of 17,013 circRNAs were identified, of which 681 circRNAs were differentially expressed. Of these DE circRNAs, 579 were u p- regulated and 103 were down regulated in calf and bull testis ( 36 ). In consistent with previous reports, we identified that most of the circRNAs were derived from exonic regions, then intergenic and intronic regions, respectively.…”

“…Testing of some artificially synthesized circular RNAs revealed that they contain an internal ribosome entry site. These circular RNAs may contain functions that encode proteins [11].…”

“…CircRNAs are a prominent new class of closed-loop RNA molecules found in eukaryotes, that are produced by the alternate shearing of precursor mRNAs ( 7 ), and are involved in the regulation of mammalian development ( 8 ), growth ( 9 ), reproduction ( 10 ), and immunity ( 11 ) in animals. For example, circPPARA can act as an endogenous sponge to adsorb mir-429 and mir-200b , thereby promoting intramuscular adipogenesis in pigs ( 12 ), and reduced expression of circZNF609 significantly inhibits myoblast proliferation in mice in vivo ( 13 ).…”
