Circular RNAs in Cardiovascular Diseases: Regulation and Therapeutic Applications

Abstract: Cardiovascular disease is one of the leading causes of mortality worldwide. Recent studies have shown that circular RNAs (circRNAs) have emerged as important players in the prevention and treatment of cardiovascular diseases. circRNAs are a class of endogenous noncoding RNAs that are generated by back-splicing and are involved in many pathophysiological processes. In this review, we outline the current research progress on the regulatory roles of circRNAs in cardiovascular diseases. Further, new technologies a… Show more

“…circRNA is formed when a downstream 5’ splice site (splice donor) in a pre-mRNA molecule joins with an upstream 3’ splice site (splice acceptor) in the same molecule through a noncanonical splicing event. This results in the formation of a covalently closed loop structure 80 , 82 , 83 . In the case of circSKA3, it would be generated from the SKA3 pre-mRNA, where specific exons or introns are back-spliced together to form the circular structure.…”

“…The use of circRNAs as therapeutic targets in CAD has been widely discussed [ 24 , 29 , 170 ]. An attractive therapy strategy is the creation and use of artificial circRNAs in the form of miRNA sponges [ 171 ].…”

“…An attractive therapy strategy is the creation and use of artificial circRNAs in the form of miRNA sponges [ 171 ]. The advantages and disadvantages of the methodology of circRNA synthesis in vitro have been thoroughly reviewed recently [ 170 ]. The use of newly synthesized circRNAs is restricted by imperfect cyclization, the presence of extraneous fragments, their off-target effects, and the activation of the immune response.…”

“…For example, cir-znf215 has been found to promote the growth and metastasis of cholangiocarcinoma by inhibiting the AKT pathway ( Liao et al, 2023 ). circRNAs also exhibit tissue- and cell-specific expression ( Wang et al, 2023 ). Therefore, circRNAs could potentially serve as diagnostic and prognostic markers as well as therapeutic targets for PCa.…”
