Circulating levels of adrenomedullin in patients with Addison's disease before and after corticosteroid treatment

Abstract: BACKGROUND AND OBJECTIVE A novel hypotensive peptide, adrenomedullin (AM), had recently been isolated, purifed and its encoding gene sequenced from a human phaeochromocytoma. In humans AM circulates in physiological levels and possesses a distinct vasodilatatory activity. The purpose of this study was to examine the behaviour of AM levels in primary adrenal failure. DESIGN Plasma AM levels were measured in patients with proven Addison's disease using a specific radioimmunoassay and compared to those in healthy… Show more

“…AM was measured by a specific radioimmunoassay (RIA, Phoenix Pharm Inc., Mountain View, CA, USA) as previously described 15 . The anti-AM antibody crossreacts 100% with human AM-(1-52) but not with rat AM-(1-50), human amylin, human calcitonin gene-related peptide, endothelin-1, a-atrial natriuretic peptide (1-28), and ~-atrial natriuretic peptide.…”

“…AM was measured with a specific radioimmunoassay (RIA, PHOENIX PHARM; Inc. Mountain View, CA; USA) as reported (12). The anti-AM antibody cross-reacts at 100% with human AM-( 1-52) but not with rat AM-( 1-50), human amilin, human calcitonin gene related peptide, endothelin-1, a. atrial natriuretic peptide-( 1-28),~atrial natriuretic peptide- (32) or catrial natriuretic peptide- (22).…”

“…For example, we do not know how pancreatic AM production is controlled. Glucocorticoids or atrial natriuretic peptide, which promote a decrease and an increase in AM circulating levels, respectively (Letizia et al, 1998;Vesely et al, 1996), may be involved. The vasodilatory effect of AM may also have some influence on insulin secretion by elevating the pancreatic perfusion rate, but this remains to be established in vivo.…”
