Class I major histocompatibility complex cDNA clones from sheep thymus: alternative splicing could make a long cytoplasmic tail

Abstract: To investigate the class I major histocompatibility complex (MHC) genes expressed in the young sheep thymus, a cDNA library was screened with a human HLA-B7 cDNA probe under conditions of relaxed stringency. Thirteen clones were isolated and found by partial sequences to fall into five classes, requiring the expression of at least three loci. One sequence was found six times, almost half of the total, and may thus represent the major message expressed in the young sheep thymus. One of the clones was found to h… Show more

“…The partial cDNA, (Figs. This replacement is atypical of functional MHC class I molecules and is not found in other artiodactyls (Garber et al 1993;Grossberger et al 1990), rats (Rada et al 1990 based upon d (Jukes and Cantor 1969), using the method of Rzhetsky and Nei (1992). The aliphatic amino acid leucine 294 in the transmembrahe region was replaced with the basic amino acid histidine (Fig.…”

“…Mixed bases in A3iiH3 are: M = A or C, S = C or G, R = A or G. Artiodactyl MHC class I sequences compared were: cattle: BL3-6 and BL3-7 ); pBolA1 (Brown et al 1989); pCAI10 (Grossberger et al 1990). The PCR primer A3iiH3 had three mixed bases, as indicated below.…”

“…Polymorphic restriction fragments hybridized with the class I and class II probes and could be correlated with MHC serotypes. Moreover, Grossberger et al . (1990) proposed the expression of at least three class I loci on the basis of the DNA sequences of 13 cDNA clones from a lamb thymus.…”
