Class II-restricted presentation of an endogenously derived immunodominant T-cell determinant of hen egg lysozyme.

Abstract: An in vitro model was used to investigate the potential for different structural forms of endogenous antigen to be processed and presented by major histocompatibility complex class H molecules. For this purpose the class IIrestricted presentation of an immunodominant epitope of hen egg lysozyme Mixing experiments with different antigenpresenting cells indicated that the HEL-(46-61) determinant was derived from endogenous antigen rather than by reuptake of shed or secreted HEL determinants. We conclude that MI… Show more

“…As demonstrated by flow cytometry, each transfectant expressed surface mHEL recognizable with a polyclonal rabbit antisera reactive to native HEL, and also with a conformationally dependent anti-HEL mAb, F10.6.6 (20) (data not shown). We found the endogenously synthesized mHEL to be efficiently processed for presentation on MHC class II, in agreement with previous work by Brooks et al (21) performed with a similar mHEL construct.…”

“…Because of the requirement for all three plasmids in the DNA immunization, it is most likely that the cells expressing CD1 from the injected DNA also are expressing Ova and are presenting it. For class II molecules, there are ample precedents for the presentation of peptides derived from endogenously expressed, secreted proteins (34), and, therefore, it is not so surprising that peptides derived from such proteins also reach endosomes containing CD1.…”
