Click Chemistry Immobilization of Antibodies on Polymer Coated Gold Nanoparticles

Abstract: The goal of this work is to develop an innovative approach for the coating of gold nanoparticles (AuNPs) with a synthetic functional copolymer. This stable coating with a thickness of few nanometers provides, at the same time, stabilization and functionalization of the particles. The polymeric coating consists of a backbone of polydimethylacrylamide (DMA) functionalized with an alkyne monomer that allows the binding of azido modified molecules by Cu(I)-catalyzed azide/alkyne 1,3-dipolar cycloaddition (CuAAC, c… Show more

“…Silica or silica-coated particles with Si-OH groups on their surface were grafted with trialkoxysilanes with amine groups [ 214 , 215 ] and polymers containing trihydroxysilane moieties and clickable alkyne functions [ 216 ]. The particles with amine functions were further modified with 2-bromoisobutyryl bromide, yielding particles with ATRP initiator and/or with lanthanide-doped 4-cyano-4-((dodecylsulfanylthiocarbonyl) sulfanyl) pentanoic acid-produced particles with chain-transfer agent used in RAFT polymerizations.…”

“…For example, Finetti et al successfully immobilized an azido modified anti-mouse IgG antibody on alkyne-functionalized gold nanoparticles via CuAAC click chemistry. [117] Due to the cytotoxicity of copper(I) catalyst, some copper-free click reactions have been also developed, such as strain-promoted azide-alkyne cycloaddition (SPAAC) where the alkyne part is a highly strained cyclic alkyne ( e.g. cyclooctyne).…”

“…SeNPs also show favourable biocompatibility and can be easily conjugated with biomolecules without losing activity and preparing them is easy and cost effective ( Wang et al, 2019 ). Biomolecules such as antibodies can be immobilized on both AuNCs and SeNPs either passively through ionic or hydrophobic interactions or covalently through a chemical reaction with a surface group ( Finetti et al, 2016 ).…”
