Clinical, cytogenetic and molecular genetic characterization of a tandem fusion translocation in a male Holstein cattle with congenital hypospadias and a ventricular septal defect

Abstract: Hypospadias, disorder of sex development (DSD), is a sporadic congenital abnormality of the genital region in male ruminants, which is characterized by a non-fused urethra during fetal development. Detailed clinical examination classified the hypospadias phenotype of a male Holstein calf studied here as the perineal type. In combined use of cytogenetic analysis and whole genome sequencing, a non-mosaic, pseudo-monosomy 59, XY + tan(18;27) was detected. This chromosomal aberration had its origin in a tandem fus… Show more

“…Two cases of trisomy involving BTA20 and BTA29 have been found using only genomic analyses [ 30 , 40 ]. It should be interesting to compare this approach with cytogenetic analyses, such as chromosome banding and FISH mapping using specific chromosome markers, as recently performed in a case of tandem fusion translocation [ 42 ]. A useful approach to detecting numerical chromosome abnormalities using a FISH-mapping technique with two marker chromosomes has been applied to cattle embryos derived from in vitro production (IVP) [ 43 ].…”

“…Hypospadias were also found in other domestic animals such as dogs, 122 cats, 123 sheep, 124 cattle, 125 and horses. 126 Moreover, polymorphisms were also found in MAMLD1 , SRD5A2 , and AR genes in hypospadias dogs.…”

“…15 A literature search suggests that the rabbit model is more commonly used for hypospadias repair studies, rather than for etiologic research of hypospadias. 121 Hypospadias were also found in other domestic animals such as dogs, 122 cats, 123 sheep, 124 cattle, 125 and horses. 126 Moreover, polymorphisms were also found in MAMLD1, SRD5A2, and AR genes in hypospadias dogs.…”
