Clinical Evaluation of the ASTY Colorimetric Microdilution Panel for Antifungal Susceptibility Testing

Abstract: A method using a commercially prepared colorimetric microdilution panel (ASTY; Kyokuto Pharmaceutical Industrial Co., Ltd.) was compared in four different laboratories with the National Committee for Clinical Laboratory Standards (NCCLS) reference microdilution method by testing 802 clinical isolates of Candida spp. (C. albicans, C. glabrata, C. tropicalis,C. parapsilosis, C. krusei, C. lusitaniae, C. guilliermondii, C. lipolytica, C. rugosa, and C. zeylanoides) against amphotericin B, 5-fluorocytosine (5FC), … Show more

“…(C. albicans, C. glabrata, C. tropicalis, C. parapsilosis, C. krusei, C. lusitaniae, C. guilliermondii, C. lipolytica, C. rugosa and C. zeylanoides) against amphotericin B, 5-fluorocytosine, fluconazole and itraconazole. 62 The ASTY colorimetric microdilution panel method appears to be comparable with the CLSI method for testing the susceptibility of Candida spp. to a variety of antifungal agents.…”

“…The results suggest that the Sensititre and ASTY colorimetric broth microdilution methods are suitable alternatives for routine antifungal susceptibility testing of Candida spp. and C. neoformans, showing excellent agreement (Ն80%) with the NCCLS reference method for amphotericin B, flucytosine, fluconazole, and itraconazole (3,12,20). The FUNG-ITEST breakpoint testing system is a simplified method in which each drug (amphotericin B, flucytosine, fluconazole, ketoconazole, and itraconazole) is tested at only two concentrations to distinguish resistant isolates from susceptible ones.…”

“…recovered during prolonged treatment with this agent correlate with therapeutic failure (8,11). Previous evaluations of commercially available antifungal drug susceptibility test methods have included colorimetric broth microdilution methods (Sensititre, Westlake, Ohio, and ASTY, Kyokuto Pharmaceutical Industrial Co., Ltd.) (3,12,20), a noncolorimetric broth breakpoint testing system (FUN-GITEST; Sanofi Diagnostics Pasteur, Paris, France) (2,21), and an agar diffusion system (E-test; AB Biodisk, Solna, Sweden) (4,23). The results suggest that the Sensititre and ASTY colorimetric broth microdilution methods are suitable alternatives for routine antifungal susceptibility testing of Candida spp.…”

“…(Carrillo-Munoz et al, 2006) ASTY colorimetric microdilution panel is another method, which is under investigation. (Pfaller et al, 1998) . Further, there is another non-commercial method which is based on mitochondrial dehydrogenase enzymes reducing tetrazolium salts 2,3-bis {2-methoxy-4-nitro-5-[(sulfonyl amino) carbonyl]-2H-tetrazolium-hydroxide} (XTT).…”
