Clinical evaluation of total and high-avidity anti-dsDNA antibody assays for the diagnosis of systemic lupus erythematosus

Abstract: Background This study evaluated the diagnostic performances of total and high-avidity (HA) anti-dsDNA enzyme immunoassays (EIA) in Chinese systemic lupus erythematosus (SLE) patients. Methods A total of 410 serum samples from 217 SLE patients, 54 patients with other systemic autoimmune diseases, and 139 healthy subjects were tested on total and HA anti-dsDNA EIA, as well as three commercial in vitro diagnostic kits: BioPlex 2200 ANA Screen, Kallestad anti-dsDNA EIA, and Crithidia Lucilae IFA. The disease activ… Show more

“…Similar to the previous studies, miR-101-3p expression was markedly reduced in SLE in the current study. To date, SLE diagnosis is supported by a variety of indices including anti-dsDNA, ESR, CRP, IgG and C3 (33)(34)(35)(36). Reduced expression of miR-98 is found to be associated with anti-dsDNA and SLEDAI in SLE PBMCs (37) while miR-203 expression is negatively associated with ESR and CRP in patients with SLE (38).…”

“…On the other hand, the avidity of the antigen-antibody interaction is also associated with the severity of autoimmune diseases [77,78]. In addition, avidity is a parameter that directly influences immunodiagnostic reactions, including avidity measurements being used to assess the stage of a given pathology [79][80][81][82][83].…”

“…Antidouble-stranded DNA (anti-dsDNA) autoantibodies play a pivotal role in the diagnosis and follow-up of SLE [1][2][3] and are related to the development of lupus nephritis. 4 5 The 2019 European League Against Rheumatism (EULAR) and the American College of Rheumatology (ACR) criteria for classifying SLE include positive values for SLE-specific antibodies, such as anti-dsDNA autoantibodies, as the criterion with the highest weight in the immunology domains.…”
