Clinical relevance of 13 cytokine gene polymorphisms in Chinese major trauma patients

Abstract: Polymorphisms of IL-1beta/-1470, IL-1beta/-511, IL-1beta/-31, IL-4/-589, IL-6/-572, IL-8/-251, IL-10/-819, and TNFalpha/-308 are susceptibility loci for the development of sepsis and organ dysfunction in major trauma patients.

“…Single nucleotide polymorphisms (SNPs) in inflammatory cytokine genes such as those coding for TNF and LTA have been extensively evaluated in adults [12][13][14][15][16][17], providing good markers for severity in adult sepsis. A recent meta-analysis has confirmed an increased risk of sepsis in carriers of the A allele for the TNF-308 G > A SNP [6] and AA genotype for LTA + 252A > G SNP [7].…”

“…Single-nucleotide polymorphisms of nine cytokine genes (out of 13) were associated with ex vivo cytokine production by LPS-stimulated leukocytes or risk of development of sepsis in a cohort of trauma patients [39]. A relationship between IL-6, IL-10 and the responsiveness to an ACTH stimulation test were also reported by Kwon et al [40], strongly confirming an interference between the inflammatory response and the neuro-endocrine pathway.…”

“…Only two studies [15,17] with high quality containing 193 cases and 193 controls evaluated the association of the IL-1A-889 polymorphism (rs1800587) with sepsis risk. In overall comparison, a significant association was observed for allelic effect (OR = 1.47, 95% CI = 1.01-2.13, P = 0.04).…”

“…Seven studies [12-15,17,34] totaling 745 cases and 750 controls were identified in order to investigate the association between IL-1B-511 polymorphism (rs16944) and sepsis risk. Overall, the results showed no associations under any genetic model.…”
