Clinical relevance of transforming growth factor α, epidermal growth factor receptor, p53, and Ki67 in colorectal liver metastases and corresponding primary tumors

Abstract: To determine whether the expression of transforming growth factor ␣ (TGF-␣), its receptor (epidermal growth factor receptor [EGFr]), p53 nuclear protein, and proliferation influences prognosis of patients with liver metastases, a study was performed in 45 liver metastases and 33 corresponding primary colorectal carcinomas in patients referred for liver surgery. The expression of TGF-␣, EGFr, p53 nuclear protein, and proliferation rate was correlated with clinicopathological characteristics and survival after p… Show more

“…Another aspect of the cytokine hypothesis is guided by the emergence of an understanding of the role of TGF-a in cancer prognosis (30) and signaling within the central nervous system (31 -35). TGF-a is a ligand for the EGF receptor (EGFR1) and colorectal cancer patients having both elevated tumor EGFR expression and increased circulating levels of TGF-a have relatively poor prognosis (23,36). In our study, we show a significant correlation between symptoms, poor QOL, worsened appetite loss, and perhaps most significantly, increased fatigue in the patients with elevated serum levels of TGF-a.…”

“…[13][14][15][16] In contrast, an inverse association was observed with cervical cancer prognosis 17 and no association was found with prognosis in patients with colon and pancreatic cancer. 18,19 In a study of patients undergoing surgical resection for hepatocellular carcinoma, higher levels of expression of Ki-67 in tumor tissue were associated with higher tumor grade 20 and early disease recurrence. 3 Identification of additional features of hepatocellular carcinoma that are associated with rapid posttransplant recurrence could be used to improve patient selection for transplantation and to differentiate those who could benefit from closer surveillance or adjuvant therapy.…”

“…14,15 In a study of 45 liver metastases and 33 corresponding primary colorectal carcinomas, the absence of EGFR expression was associated with a better survival after partial liver resection. 16 Therefore, we investigated the correlation between survival and immunohistochemical expression of VEGF by tumor cells as well as TAM/stroma, and of EGFR by tumor cells in primary colon carcinoma specimens.…”
