Clinical significance of microRNA-183/Ezrin axis in judging the prognosis of patients with osteosarcoma

Abstract: MicroRNA-183 (miR-183) has been demonstrated to be a tumor suppressor miRNA in inhibition of migration and invasion of osteosarcoma by targeting an oncogene Ezrin. The aim of the present study was to determine the correlation of combined miR-183 and Ezrin mRNA expression with clinicopathologic features and prognosis in patients with osteosarcomas. Expressions of MiR-183 and Ezrin mRNA were both examined using quantitative real-time reverse transcriptase-polymerase chain reaction analysis in 68 specimens from p… Show more

“…Leonard et al [24] reported that the ezrin gene was highly expressed in human primary osteosarcoma samples and could be considered the new prognostic marker in osteosarcoma. Mu et al [25] proved that ezrin mRNA expression was higher in osteosarcoma than in noncancerous bone tissues, and high ezrin expression was also significantly correlated with the high grade of the tumour. Data presented by Salas et al [26] suggested that ezrin is a useful immunohistochemical marker for differential diagnosis between chondroblastic osteosarcomas and conventional chondrosarcomas, with a specificity of 100 %.…”

“…In pediatric cancers, osteosarcoma accounts for 8 % incidence rate and is a leading cause of cancer-related death among young adults [3,4]. The etiology of osteosarcoma involves environment factors and genetic predisposition [5,6].…”

“…Moreover, the combined miR-183 downregulation and ezrin upregulation show a positive correlation with high tumor grade, poor response to chemotherapy, positive metastasis and recurrence, and lower five-year survival rate [48]. However, there is no significant association between miR-183/ezrin expression and other clinicopathological parameters, including patients' age, sex, site, histologic type and surgical method [48]. It has been reported that miR-183 mimics markedly diminish the migratory and invasive abilities of F5M2, a high-metastatic human osteosarcoma cell line, by targeting ezrin [49].…”

“…More recently, Mu et al analyzed the correlation of combined miR-183 and ezrin mRNA expression with clinicopathologic features and prognosis in patients with OS. They found that miR-183 expression is significantly decreased but ezrin mRNA expression is increased in specimens derived from OS cases compared with corresponding noncancerous bone tissues [48]. Moreover, the combined miR-183 downregulation and ezrin upregulation show a positive correlation with high tumor grade, poor response to chemotherapy, positive metastasis and recurrence, and lower five-year survival rate [48].…”
