Clobetasone butyrate, a new topical corticosteroid: clinical activity and effects on pituitary-adrenal axis function and model of epidermal atrophy.

Abstract: Clobetasone butyrate is a new corticosteroid, selected for study because of its combitation of good activity in the vasoconstriction test and low systemic activity in animals. Formulated as an 005o% ointment and cream (MolivateN it was clinically effective in patients with eczema, its activity being significantly greater than that of hydrocortisone 1% or fluocortolone 020°, (Ultradil). Under conditions that predispose to maximal percutaneous absorption clobetasone butyrate ointment had minimal effect on hypoth… Show more

“…The topical steroid clobetasone butyrate (CB) causes less systemic effect than BV when applied to the skin [11] and has been shown to be effective in reducing the deleterious effect of gluten on coeliac mucosa in vitro [12]. We now report a clinical trial of topical corticosteroids in 10 patients with sub-total villous atrophy, 7 of whom took CB and 3 BV.…”

“…In our study we chose clobetasol propionate as the strong preparation, because of its ability to rapidly resolve conditions, and clobetasone butyrate as the mild one, because of its reasonable efficacy combined with a low propensity to cause side-effects [12,17]. In many conditions it will be appro priate to all other combinations of steroid preparations with different strengths and properties.…”

“…Clobetasone butyrate, unlike betamethasone phosphate and prednisolone phosphate, is a steroid with a high cutaneous topical activity, as has been shown by its successful use in the treatment of inflammatory dermatoses (Munro and Wilson, 1975). It was postulated that because of this superior topical activity it might prove to be effective in the local treatment of external eye diseases.…”
