Cloning and Characterization of the BcTuR3 Gene Related to Resistance to Turnip Mosaic Virus (TuMV) from Non-heading Chinese Cabbage

Abstract: A cDNA clone for the turnip mosaic virus (TuMV)-induced gene in non-heading Chinese cabbage (Brassica campestris ssp. chinensis Makino cv. Duanbaigeng) was isolated and characterized. The full-length cDNA clone, designated BcTuR3, was isolated during resistance response to TuMV.

“…Real-time PCR has been used for gene expression study in many species (Chai et al 2010;Ma et al 2010;Qi et al 2010). The relative expression of AcSERK1 was measured using Thunderbird SYBR qPCR Mix (Toyobo) performed on the iQ5 real-time PCR System (Biorad).…”

“…For instance, three dominant genes named RTM1, RTM2, and RTM3 are involved in restriction of tobacco etch virus in A. thaliana (Decroocq et al, 2009). A number of typical dominant NBS-LRR resistance genes conferring resistance to potyvirus have been cloned, such as BcTuR3 related to resistance to turnip mosaic virus (TuMV) in non-heading Chinese cabbage (Ma et al, 2010), the Rsv1 gene of soybean invoking extreme resistance against soybean mosaic virus (Hajimorad and Hill, 2001), and I gene displaying resistance to bean common mosaic potyvirus (BCMV) in Phaseolus vulgaris (Kelly, 1997). To our knowledge, this is the first report that an h-type thioredoxin, acting as a molecular chaperone, is involved in combatting potyviral disease in plant.…”
