Cloning and complete nucleotide sequence of a full-length cDNA encoding a catalytically functional tumor-associated aldehyde dehydrogenase.

Abstract: To study the mechanism(s) controlling expression of the tumor-associated aldehyde dehydrogenase (tumor ALDH), which appears during rat hepatocarcinogenesis, cDNAs encoding this isozyme were cloned and identified with an antibody probe. Poly(A)-containing RNA from HTC rat hepatoma cells, which have been shown to possess high levels of tumor ALDH, was used as template to synthesize double-stranded cDNA. The cDNA was methylated to protect internal sites. Two different synthetic DNA linkers were added sequentially… Show more

“…The result is an Ah receptor-mediated positive transcriptional activation of each of the genes in the [Ah] battery (16,(26)(27)(28)(29). For some of these genes, a component of posttranscriptional regulation may also be present, such as has been characterized most completely for the mouse CYP1A2 gene (26).…”

“…Only a small percentage of preneoplastic nodules actually proceed to frank malignancy. Interestingly, the CYPlAl and CYP1A2 enzymes are undetectable, while the Nmo-1, Aldh-1, Ugt-1, and Gt-1 enzyme activities in the preneoplastic nodules are elevated (16,27,29,50,51). It appears likely that these previous results with rat liver preneoplastic nodules are related to our recent observations that the murine [Ah] battery Phase II gene regulation is independent of the [Ah] battery Phase I gene regulation.…”

“…Unlike the liver enzymes, the yeast enzyme is activated by K+ ions (4, 32). Whereas the mammalian enzymes have been sequenced at the cDNA (16,19,29,34) as well as the protein (24, 33, 60) level, no sequence work has been reported for the yeast enzyme. One of our interests was to study the subcellular localization of acetaldehyde metabolism in S. cerevisiae. In the mammalian liver tissue, this was studied by selectively inhibiting the cytosolic or the mitochondrial isozyme of ALDH (11,52).…”

“…Recently, an aldehyde dehydrogenase from Aspergilltis niduluns [54] and one from rat tumor [55] were sequenced, being just 56% and 29% identical, respectively, to the mitochondrial enzymes. Of the above residues Cys-49 and Glu-268 were absent in the enzyme from A .…”
