Clostridium hiranonis sp. nov., a human intestinal bacterium with bile acid 7alpha-dehydroxylating activity.

Abstract: The Clostridium-like organisms TO-931 T and HD-17, isolated from human faeces, have high levels of bile acid 7α-dehydroxylating activity. Sequencing of their 16S rDNA demonstrated that they belong to cluster XI of the genus Clostridium and that they represent a new and distinct line of descent. Clostridium bifermentans and Clostridium sordellii in cluster XI also possess bile acid 7α-dehydroxylating activity. DNA-DNA hybridization experiments with the isolates, TO-931 T and HD-17, and C. bifermentans and C. so… Show more

“…This species belongs to Clostridium cluster XI which also includes C. difficile. 39 The latter is known to increase in relative abundance during broad-spectrum AB therapy, sometimes leading to AB-associated diarrhea and pseudomembranous colitis. 10,40,41 In this study, C. hiranonis decreased in relative abundance due to AB treatment (mean www.tandfonline.comdecrease from 0.29% to 0.01% at the DNA level).…”

“…The phylogenetic position of the two strains among Bacteroides species and representative members of the genera Prevotella, Bacteroides, Porphyromonas and Tannerella are shown in Fig. 1 (Kitahara et al, 2001) after incubation for 12 h at 37 u C, and purified by using the methods of Saito & Miura (1963). Cells were disrupted with lysozyme (Wako Pure Chemical Industries) and SDS, extracted with phenol, and the DNA was recovered using alcohol precipitation and suspended in TE buffer (1 mM EDTA, 10 mM Tris/HCl, pH 8?0).…”

“…T and 176 supported the affiliation of the novel strains with members of the genus Bacteroides, which have a G+C content of 40-48 mol% (Shah, 1992 T was extracted from cells harvested from EGF broth (Kitahara et al, 2001). DNA-DNA hybridization was performed by using the photobiotin-labelling method of Ezaki et al (1989), using a microplate reader (FluoroskanAscent; Labsystems).…”

“…Extraction and purification of bacterial DNA was performed according to the methods of Kitahara et al (2001) and purified by the method of Saito & Miura (1963). Strains of L. fructivorans, L. homohiochii and L. sanfranciscensis were not used in this study because the sequence similarities between the new isolates and the type strains of these three species were lower than the recommended value for species differentiation as proposed by Stackebrandt & Ebers (2006).…”

“…Levels of DNA-DNA relatedness between the new isolates and L. lindneri JCM 11027 T , and the DNA G+C contents of the isolates were determined according to methods described by Kitahara et al (2001). Extraction and purification of bacterial DNA was performed according to the methods of Kitahara et al (2001) and purified by the method of Saito & Miura (1963).…”
