Clozapine-induced cataract in a young female

Abstract: A variety of systemic drugs including corticosteroids, amiodarone and antipsychotics have been known to cause cataract formation. Typical antipsychotics such as chlorpromazine have been reported to cause cataract formation in varying rates ranging from 22% to 80%. Cataract as an adverse effect resulting from the long term use of atypical antipsychotic has rarely been mentioned in literature, and there is only a single case report of cataract formation from prolonged use of clozapine. We report a rare case of c… Show more

“…Производные фенотиазина приводят к развитию катаракты [6]. Частота помутнений хрусталика при приеме хлорпромазина составляет, по разным оценкам, от 22 до 80 % [25]. По другим данным, употребление данного препарата в дозе  300 мг/сут в течение  90 дней увеличивает риск развития катаракты в 8,8 раза (95%-ный доверительный интервал 3,1-25,1 раза) [26].…”

“…Doses as high as 800 mg/day for 2 years can produce pigmentary deposits on the lens [ 15 ]. The opacity of the lens can also occur, though rare on prolonged use of atypical antipsychotics such as Clozapine [ 16 ]. The mechanisms involved in the formation of these opacities are through photosensitizing agents like Chlorpromazine and its metabolites denature proteins, which become opacified when exposed to sunlight and are deposited in the skin, lens, and cornea [ 15 ].…”
