CO2 anesthesia on Drosophila survival in aging research

Abstract: Carbon dioxide (CO 2 ) exposure is a common method of anesthesia in studies of Drosophila melanogaster. A number of negative side effects of CO 2 anesthesia have been reported. It is not clear whether the length of CO 2 anesthesia time affects Drosophila survival in aging research. Here, we examined the potential effect of the CO 2 anesthesia time length of 10-150 min. We found that long CO 2 exposure could lead to Drosophila death, more significant in males. The longer the anesthesia time is, the longer it tak… Show more

“…Flies were repeatedly anesthetized using CO 2 to prevent voluntary and involuntary movements during image acquisition, CO 2 pulsing allowed the flies to awaken and feed between timepoints (Poinapen et al, 2017; Koyama et al, 2020; Tang et al, 2021). Since CO 2 anesthesia adversely affects fertility in flies (Shen et al, 2020) we minimized the duration of anesthesia by integrating image acquisition with CO 2 pulsing via an Arduino controller. The Arduino opened a solenoid valve on the CO 2 tank in response to a signal from the image acquisition software (Fig.…”

“…Flies were anaesthetized using carbon dioxide. The vials containing the flies were inverted and placed on a pad that has been connected to a source of carbon dioxide. The CO2 gas displaced the oxygen in the vial causing the flies to become exposed to carbon dioxide till they become sedated (Shen et al, 2020). The weight of each fly was recorded.…”

“…[11][12][13] In addition, many neurotoxicity and neurodegenerative studies that screen for the potential protective properties of biologically active compounds use D. melanogaster as the model organism. [14][15][16] D. melanogaster are ideal subjects to use because they are relatively easy to handle, they have a short lifespan (40 to 120 days), they reproduce rapidly, and 75-77% of human disease-related genes have functional orthologs in the fly genome. 14,17 Since its central nervous system operates on the same fundamental principles as those of humans, this makes them ideal subjects for studies into the nervous system.…”
