Cobalt proteins

Abstract: In the form of vitamin B 12 , cobalt plays a number of crucial roles in many biological functions. However, recent studies have provided information on the biochemistry and bioinorganic chemistry of several proteins containing cobalt in a form other than that in the corrin ring of vitamin B 12 . To date, eight noncorrin-cobalt-containing enzymes (methionine aminopeptidase, prolidase, nitrile hydratase, glucose isomerase, methylmalonyl-CoA carboxytransferase, aldehyde decarbonylase, lysine-2,3-aminomutase, and … Show more

“…However, none of these 35 metals was detected within the limits of the assay (20 ng/ml). In this respect, the isonitrile hydratase is quite different from the nitrile hydratases of Pseudomonas (27,28) and Brevibacterium (29) species, which contain tightly bound iron (1,30), and the nitrile hydratase of Rhodococcus rhodochrous J1, which contains cobalt as a prosthetic group (1,11,31).…”

“…NikR has been the subject of several studies showing both high affinity nickel binding and a change in coordination geometry on nickelNikR binding to operator DNA (52)(53)(54). Interest in bacterial nickel-binding proteins has been fueled by the realization that nickel deprivation may be a useful method of inhibiting bacterial growth or pathogenicity (55). Periplasmic binding proteins also have potential as biosensors for their respective ligands (56), and NikA has been touted a possible biosensor for nickel (34).…”

“…The mechanism by which nitriles are hydrolyzed by NHase is unknown, and nitriles are extremely resistant to hydrolysis. However, rates of hydration have been shown to increase dramatically in the presence of a Lewis acidic metal ion capable of coordinating nitriles.A number of structural models for Fe-NHase and Co-NHase containing oxidized thiolate ligands have been reported, [31][32][33][34][35][36] and in one case sulfur oxygenation was shown to influence the pK a of a coordinated water molecule. 37 Very few models containing singly oxygenated sulfurs are available, and none of these contain iron.…”
